The 30-year-old revealed his injury and removal from the card in a post on Twitter.
"I'm [out of] the UFC 135 card I hurt my shoulder and MRI is not looking good," Gamburyan wrote. "I just wanna say sorry to... my friends and fans I'll [be] back!"
The bout was to be Gamburyan's second fight since returning to the UFC with the absorption of the WEC. The former lightweight has lost two fights in a row, including his return bout against Tyson Griffin in June.
Nunes is now without an opponent for the moment as he looks to rebound from a loss in his last fight himself. After defeating Mike Brown in his UFC debut in January, Nunes was defeated by Kenny Floriain in his last fight in June at UFC 131.
UFC 135 takes place on Sept. 24 from the Pepsi Center in Denver, Colo., headlined by a Light Heavyweight Championship bout between Jon Jones and Quinton "Rampage" Jackson.
Penick's Analysis: That would have been a good fight, so it's an unfortunate loss to the card. Nunes will likely still be on the card, as they've got time to find him a replacement, but for Gamburyan it's a definite setback. Shoulder injuries can be seriously detrimental and hard to come back from, so for his sake hopefully it won't be that major of an injury.
